Bacterial inactivation, DNA damage, and faster ATP degradation induced by ultraviolet disinfection

Abstract: • Long amplicon is more effective to test DNA damage induced by UV. • ATP in bacteria does not degrade instantly but does eventually after UV exposure. • After medium pressure UV exposure, ATP degraded faster. The efficacy of ultraviolet (UV) disinfection has been validated in numerous studies by using culture-based methods. However, the discovery of viable but non-culturable bacteria has necessitated the investigation of UV disinfection based on bacterial viability parameters. We used quantitative polymerase chain reaction (qPCR) to investigate DNA damage and evaluated adenosine triphosphate (ATP) to indicate bacterial viability. The results of qPCR effectively showed the DNA damage induced by UV when using longer gene amplicons, in that sufficiently long amplicons of both 16S and gadA indicated that the UV induced DNA damages. The copy concentrations of the long amplicons of 16S and gadA decreased by 2.38 log/mL and 1.88 log/mL, respectively, after exposure to 40 mJ/cm2 low-pressure UV. After UV exposure, the ATP level in the bacteria did not decrease instantly. Instead it decreased gradually at a rate that was positively related to the UV fluence. For low-pressure UV, this rate of decrease was slow, but for medium pressure UV, this rate of decrease was relatively high when the UV fluence reached 40 mJ/cm2. At the same UV fluence, the ATP level in the bacteria decreased at a faster rate after exposure to medium-pressure UV.

Frontiers of Structural and Civil Engineering 2020, Volume 14, Issue 3,   Pages 746-759 doi: 10.1007/s11709-020-0635-2

Abstract: Carbonation of concrete is a process which absorbs carbon dioxide (CO ). Recycled aggregate concrete (RAC) may own greater potential in CO uptake due to the faster carbonation rate than natural aggregate concrete (NAC). A quantitative model was employed to predict the CO uptake of RAC in this study. The carbonation of RAC and the specific surface area of recycled coarse aggregates (RCAs) were tested to verify accuracy of the quantitative model. Based on the verified model, results show that the CO uptake capacity increases with the increase of RCA replacement percentage. The CO uptake amount of 1 m C30 RAC within 50 years is 10.6, 13.8, 17.2, and 22.4 kg when the RCA replacement percentage is 30%, 50%, 70%, and 100%, respectively. The CO uptake by RCAs is remarkable and reaches 35.8%–64.3% of the total CO uptake by RAC when the RCA storage time being 30 days. Considering the fact that the amount of old hardened cement paste in RCAs is limited, there is an upper limit for the CO uptake of RCAs.

A spectrally selective surface structure for combined photothermic conversion and radiative sky cooling

Bin ZHAO, Xianze AO, Nuo CHEN, Qingdong XUAN, Mingke HU, Gang PEI

Frontiers in Energy 2020, Volume 14, Issue 4,   Pages 882-888 doi: 10.1007/s11708-020-0694-z

Abstract: The sun and outer space are the ultimate heat and cold sources for the earth, respectively. They have significant potential for renewable energy harvesting. In this paper, a spectrally selective surface structure that has a planar polydimethylsiloxane layer covering a solar absorber is conceptually proposed and optically designed for the combination of photothermic conversion (PT) and nighttime radiative sky cooling (RC). An optical simulation is conducted whose result shows that the designed surface structure (i.e., PT-RC surface structure) has a strong solar absorption coefficient of 0.92 and simultaneously emits as a mid-infrared spectral-selective emitter with an average emissivity of 0.84 within the atmospheric window. A thermal analysis prediction reveals that the designed PT-RC surface structure can be heated to 79.1°C higher than the ambient temperature in the daytime and passively cooled below the ambient temperature of approximately 10°C in the nighttime, indicating that the designed PT-RC surface structure has the potential for integrated PT conversion and nighttime RC utilization.
